New Approaches Aim to Regrow Joints Damaged by Arthritis

Scientists are exploring innovative ways to repair joints affected by osteoarthritis, raising hopes for more effective treatments. The Advanced Research Projects Agency for Health (ARPA-H) reports that several experimental approaches may help regenerate damaged cartilage and bone.

Osteoarthritis, which affects around 32 million people in the United States, develops as joint tissues gradually wear down over time. Current treatments mainly relieve symptoms, but do not restore the damaged structures.

Three research teams are focusing on new solutions, particularly for knee osteoarthritis. Two groups—based at Duke University and the University of Colorado Boulder—are developing injectable or infusion-based therapies designed to stimulate the regrowth of cartilage and bone.

A third team at Columbia University is working on a more ambitious concept: regenerating an entire knee joint using a 3D-printed scaffold populated with bone and cartilage cells.

So far, these techniques have only been tested in small animal models, and results may differ in humans. However, some methods have already demonstrated the ability to restore cartilage within a few months.

One approach involves a drug that is gradually released inside the joint to promote tissue repair over time. Another uses proteins that fill damaged areas and trigger natural cartilage regeneration.

Experts say these developments could represent a major breakthrough. Unlike current therapies, which focus on symptom management, these methods aim to address the root cause by rebuilding damaged tissue. Researchers are now working to harness the body’s own regenerative capacity to restore joint function.

Clinical trials in humans are expected to begin within the next 18 months. The program has received significant funding, with tens of millions of dollars invested to accelerate progress. If successful, these treatments could not only transform arthritis care but also be made more affordable for patients.
